Abstract

Zingiberaceae merupakan salah satu genus yang paling banyak belum teridentifikasi di Kebun Raya Bogor. Total 89 genus dari famili ini diketahui belum terkonfirmasi, salah satunya Globba. Kurang kuatnya data morfologi yang sering dipengaruhi oleh faktor lingkungan menjadikan penelitian Globba secara molekuler penting untuk dilakukan. Penelitian ini bertujuan untuk mengidentifikasi ciri karakter morfologi Globba sp. (Zingiberaceae) Asal Sumatra Utara koleksi Kebun Raya Bogor, menganalisis penggunaan metode DNA barcoding region Internal Transcribed Spacer (ITS) untuk mendeterminasi Globba sp. secara molekuler, dan menganalisis kekerabatan filogenetik Globba sp. (Zingiberaceae) asal Sumatra Utara koleksi Kebun Raya Bogor menggunakan DNA barcoding region Internal Transcribed Spacer (ITS).
Hasil penelitian menunjukan sekuen ITS dari Globba sp. memiliki karakteristik berupa panjang sekuen 807 bp dengan persentase nukleotida T sebanyak 23,9%, nukleotida C 22,3%, nukleotida A 21,3 % dan nukleotida G sebanyak 32,5%. Hasil pohon kekerabatan menggunakan metode statistik UPGMA dan Neighbor Joining dengan nilai bootstrap 1000 menunjukan Globba sp. berkerabat dengan G. schomburgkii, G. sessiliflora dan G. marantina yang mana hasil tersebut diperkuat dengan analisis ABGD (Automatic Barcode Gap Discovery). Hasil analisis pohon filogenetik menunjukan bahwa Globba sp. memiliki hubungan kekerabatan paling dekat dengan G. schomburgkii (KJ872143.1) dan berkerabatan paling jauh dengan G. marantina (KX065412.1). Nilai jarak genetik terendah (0,003) dimiliki oleh Globba variabilis sedangkan nilai tertinggi (0,102) dimiliki oleh sekuen Alpinia japonica. Hasil analisis DNA barcoding menggunaan sekuen region ITS berhasil mendeterminasi spesies Globba sp.

Abstract

Zingiberaceae is one of the most unidentified genera in the Bogor Botanical Gardens. A total of 89 genera of this family are known to be unconfirmed, one of which is Globba. The lack of strong morphological data which is often influenced by environmental factors makes Globba molecular research important to do. This study aims to identify the morphological characteristics of Globba sp. (Zingiberaceae) from North Sumatra from the Bogor Botanical Gardens collection, analyzed the use of the DNA barcoding region Internal Transcribed Spacer (ITS) method to determine Globba sp., and analyzing the phylogenetic relationship of Globba sp. (Zingiberaceae) from North Sumatra from the Bogor Botanical Gardens collection using DNA barcode region Internal Transcribed Spacer (ITS).
The results showed that the ITS sequence from Globba sp. has the characteristics of a sequence length of 807 bp with a percentage of T nucleotides as much as 23.9%, C nucleotides 22.3%, A nucleotides 21.3% and G nucleotides 32.5%. The results of the phylogenetic tree using the UPGMA and Neighbor Joining statistical methods with a bootstrap value of 1000 show Globba sp. related to G. schomburgkii, G. sessiliflora and G. marantina which results were confirmed by ABGD (Automatic Barcode Gap Discovery) analysis.The phylogenetic tree analysis showed that Globba sp. has the closest relationship with G. schomburgkii (KJ872143.1) and has the most distant relationship with G. marantina (KX065412.1). The lowest genetic distance value (0.003) was owned by G. variabilis while the highest value (0.102) was owned by the Alpinia japonica sequence. The results of DNA barcoding analysis using the ITS region sequence succeeded in determining the species Globba sp.
