Abstract

Cercestis mirabilis dan Anthurium pentaphyllum merupakan jenis tanaman koleksi Kebun Raya Bogor yang berbeda. Kedua tanaman ini memiliki karakter dan bentuk morfologi yang mirip, sehingga diperkirakan terdapat kerancuan pada penamaan jenis tanaman. Identifikasi jenis secara morfologi dan barcoding dilakukan sebagai upaya mengungkapkan keanekaragaman tumbuhan. Salah satu region yang digunakan dalam barcoding yaitu trnL-trnF Intergenic Spacer. Penelitian ini bertujuan untuk menganalisis perbandingan ciri morfologi dan mengetahui hasil identifikasi C. mirabilis dan komparasinya dengan A. pentaphyllum berdasarkan sekuen trnL-trnF IGS. Data hasil isolasi DNA C. mirabilis dan A. pentaphyllum koleksi Kebun Raya Bogor sesuai dengan prosedur kit TIANGEN. Tahapannya dimulai dari amplifikasi PCR dilanjutkan elektroforesis dan sekuensing 1st BASE Singapura. Analisis data dilakukan menggunakan software Molecular Evolutionary Genetics Analysis (MEGA 11). Alignment dilakukan menggunakan ClustalW dan dianalisis menggunakan Basic Local Alignment Search Tool (BLAST) di laman National Center for Biotechnology Information (NCBI). Rekontruksi pohon filogenetik menggunakan Neighbor Joining (NJ) dengan nilai bootstrap 1000 kali. Analisis lanjut menggunakan Multiple
Sequence Alignment by Florence Corped (MultAlin). Hasil penelitian menunjukan terdapat kemiripan morfologi antara C. mirabilis dan A. pentaphyllum yang ada di KRB setelah dibandingkan dengan A. pentaphyllum yang ada di referensi. Hasil pohon filogenetik menunjukan C. mirabilis (CM KRB) adalah jenis dari A. pentaphyllum (AP KRB) setelah dikomparasi, sehingga perlu dikonfirmasi ulang bahwa terdapat kerancuan dalam penamaan tanaman di vak KRB.

Abstract
Cercestis mirabilis and Anthurium pentaphyllum are different types of plants in the Bogor Botanical Gardens collection. These two plants have similar morphological characters and forms, so it is thought that there is confusion in the naming of plant species. Identification of morphological species and barcoding was carried out as an effort to reveal plant diversity. One of the regions used in barcoding is trnL-trnF Intergenic Spacer. This study aims to analyze the comparison of morphological characteristics and determine the results of the identification of C. mirabilis and its comparison with A. pentaphyllum based on the trnL-trnF IGS sequence. Data on the results of DNA isolation of C. mirabilis and A. pentaphyllum from the Bogor Botanical Garden collection according to the TIANGEN kit procedure. The stages started with PCR amplification followed by electrophoresis and 1st BASE Singapore sequencing. Data analysis was performed using Molecular Evolutionary Genetics Analysis (MEGA 11) software. Alignment was performed using ClustalW and analyzed using the Basic Local Alignment Search Tool (BLAST) on the National Center for Biotechnology Information (NCBI) website. Phylogenetic tree reconstruction using Neighbor Joining (NJ) with a bootstrap value of 1000 times. Further analysis using Multiple Sequence Alignment by Florence Corped (MultAlin). The results showed that there were morphological similarities between C. mirabilis and A. pentaphyllum in KRB when compared with A. pentaphyllum in the reference. The results of the phylogenetic tree showed that after comparison, C. mirabilis (CM KRB) was the type of A. pentaphyllum (AP KRB), so it needed to be reconfirmed that there was confusion in the naming of plants in the KRB area.
