Abstract

Radikal bebas yang terbentuk akibat pola hidup masyarakat seperti konsumsi makanan instan dengan gizi tidak seimbang, kurangnya aktivitas fisik, kurang istirahat, merokok, dan minum minuman beralkohol akan memicu terjadinya stress oksidatif dan menimbulkan berbagai penyakit degeneratif. Antioksidan menjadi salah satu solusi untuk menghindari penyakit degeneratif tersebut. Senyawa metabolit sekunder yang termasuk antioksidan adalah fenolik. Senyawa fenolik berperan sebagai antioksidan alami dan merupakan kelompok senyawa terbesar yang ditemukan pada tumbuhan. Air Nabeez kurma Ajwa merupakan salah satu minuman kesukaan Rasulullah SAW dan memiliki potensi besar untuk dijadikan sumber antioksidan alami.
Tujuan penelitian ini untuk mengetahui kandungan total fenolik dan aktivitas antioksidan pada air Nabeez kurma Ajwa (Phoenix dactylifera L.) dengan variasi lama perendaman dan jumlah kurma. Penelitian ini merupakan penelitian eksperimen laboratorium dengan rancangan penelitian yang dilakukan yaitu RAL (Rancangan Acak Lengkap) faktorial yang terdiri dari 2 faktor, yaitu faktor lama perendaman (L) dan faktor jumlah kurma (J). Faktor pertama, lama perendaman terdiri atas 3 taraf yaitu L1 = 12 jam, L2 = 24 jam, L3 = 36 jam. Faktor kedua adalah jumlah kurma (J), terdiri atas 2 taraf yaitu J1 = 7 kurma dan J2 = 9 kurma. Terdapat 6 kombinasi perlakuan dengan 3 kali pengulangan. Metode yang digunakan yaitu analisis total fenolik dengan metode Folin-Ciocalteu dan aktivitas antioksidan dengan metode DPPH.
Hasil kadar total fenolik pada air Nabeez kurma Ajwa (Phoenix dactylifera L.) dengan variasi 7 buah 12 jam sebesar 0,772 mgGAE/gr sampel, 9 buah 12 jam sebesar 0,923 mgGAE/gr sampel, 7 buah 24 jam sebesar 1,261 mgGAE/gr sampel, 9 buah 24 jam sebesar 1,812 mgGAE/gr sampel, 7 buah 36 jam sebesar 1,726 mgGAE/gr sampel, dan 9 buah 36 jam sebesar 1,420 mgGAE/gr sampel. Hasil uji aktivitas antioksidan diperoleh hasil nilai pada air Nabeez kurma Ajwa (Phoenix dactylifera L.) dengan variasi 7 buah 12 jam sebesar 125 ppm (antioksidan sedang), 9 buah 12 jam sebesar 101 ppm (antioksidan sedang), 7 buah 24 jam sebesar 59 ppm (antioksidan kuat), 9 buah 24 jam sebesar 50,5 ppm (antioksidan kuat), 7 buah 36 jam sebesar 169 ppm (antioksidan sedang), dan 9 buah 36 jam sebesar 147 ppm (antioksidan sedang).
Kadar total fenolik dan aktivitas antioksidan tertinggi terdapat pada air Nabeez dengan variasi jumlah kurma 9 buah dan lama perendaman 24 jam yaitu total fenolik sebesar 1,812 mgGAE/gr sampel dan aktivitas antioksidan dengan nilai sebesar 50,5 ppm.

ABSTRACT
Free radicals that are formed due to people's lifestyles such as consumption of instant food with unbalanced nutrition, lack of physical activity, lack of rest, smoking, and drinking alcoholic beverages will trigger oxidative stress and cause various degenerative diseases. Antioxidants are one solution to avoid these degenerative diseases. Secondary metabolite compounds including antioxidants are phenolic. Phenolic compounds act as natural antioxidants and are the largest group of compounds found in plants. Water Nabeez Ajwa dates is one of the favorite drinks of the Prophet Muhammad and has great potential to be used as a source of natural antioxidants.
The purpose of this research is to determine the total phenolic content and antioxidant activity in Nabeez Ajwa dates (Phoenix dactylifera L.) water with variations in soaking time and the number of dates. This research is a laboratory experimental study with the research design carried out, namely the factorial completely randomized design which consists of 2 factors, namely the immersion time factor (L) and the number of dates factor (J). The first factor, the immersion time consisted of 3 levels, L1 = 12 hours, L2 = 24 hours, L3 = 36 hours. The second factor is the number of dates (J), consisting of 2 levels, J1 = 7 dates and J2 = 9 dates. There were 6 treatment combinations with 3 repetitions. The method used is total phenolic analysis using the Folin-Ciocalteu method and antioxidant activity using the DPPH method.
Total phenolic content in Nabeez Ajwa dates (Phoenix dactylifera L.) water with a variation of 7 pieces 12 hours was 0,772 mgGAE/gr sample, 9 pieces 12 hours was 0,923 mgGAE/gr sample, 7 pieces 24 hours was 1,261 mgGAE/gr sample , 9 pieces 24 hours was 1,812 mgGAE/gr sample, 7 pieces 36 hours was 1,726 mgGAE/gr sample, and 9 pieces 36 hours was 1,420 mgGAE/gr sample. The results of the antioxidant activity test showed the value of in Nabeez Ajwa dates (Phoenix dactylifera L.) water with a variation of 7 pieces 12 hours was 125 ppm (medium antioxidant), 9 pieces 12 hours was 101 ppm (medium antioxidant), 7 pieces 24 hours was 59 ppm (strong antioxidant), 9 pieces 24 hours was 50,5 ppm (strong antioxidant), 7 pieces 36 hours was 169 ppm (medium antioxidant), and 9 pieces of 36 hours was 147 ppm (medium antioxidant).
The highest levels of total phenolic and antioxidant activity were found in Nabeez water with variations in the number of dates 9 fruits and soaking time for 24 hours, total phenolic is 1,812 mgGAE/gr sample and antioxidant activity with a value of is 50,5 ppm.
